The dried blood spot method DBS ; is expected to isolate DNA with less hands on time and requiring lower transport and DNA isolation costs and therefore could be an good alternative for the commercial available DNA isolation kits. The usefulness of capillary blood obtained by fingerprick for genotyping has been described previously. However, a method for DBS for genotyping in correlation with pharmacogenomics has not yet been reported in literature. Methods: EDTA blood samples and an in vitro made blood spots on filter paper were obtained from 106 Chinese renal transplant recipients. DNA isolation from blood samples is performed by using 200 ml EDTA whole blood with a column method according to the manufacturers' instructions. For the DBS method B ; a few drops of the same EDTA blood samples is pipetted on the sampling paper. After drying for hour the analyst punches out a paper disk from the blood spot sample. Additionally, the 3 mm paper disk is transferred in to an cup, 500 ml sterile water is added and vortexed 3 times during 5 seconds. The sterile water is pipetted off. After adding 200 l 10% Chelex-100 solution, the cup is put in a water bath at 95C for 30 minutes. Finally, the 200 l DNA solution is pipetted in a new sterile cup and the DNA is ready for use. The previously described real-time PCR FRET assays for the CYP3A4 A-392G, CYP3A5 A6986G, ABCB1 C1236T, G2677T A and C3435T were used to examined the melting curves of both DNA isolation methods. Results: After analysing 106 melting curves of five different real-time PCR FRET assays identical results were obtained with the DNA extracted with column method and with the results of the DNA isolated with the blood spot procedure.
